Excellent articl - the interpolation vs extrapolation framing really nails the core problem. The RG7112 example upfront is perfect for showing how perfect target engagement means nothing when the biological system has compensatory pathways we didnnt model. I've seen similar failures in clinical pipelines where everything looked great on paper until humans revealed complexity we couldn't capture in vitro.
